With a 5-4 vote, the court granted a request by 27 states and several companies and business groups to block clean energy plan Administration.
The decision means that the regulations will not take effect while continuing litigation over its legality.
The states, led by the big coal producer, West Virginia, and oil producer, Texas, and several major business groups launched several lawsuits industry last October to stop the president's initiative.
More than a dozen other states and the National League of Cities, which represents more than 19,000 US cities, filed court documents supporting the Government and the Agency for Environmental Protection (EPA).
However, this decision of the Supreme is not definitive about the regulations, since the case is pending the verdict of a court of appeals.
Still, it is likely that after the verdict the case back to the Supreme rise and today's decision is a sign of skepticism from the judges on the regulation.
The appeals court has yet to hear oral arguments on June 2 and decide whether the regulations are lawful.
